제품설명

Lenti-X™ qRT-PCR Titration Kit는 viral RNA 정제를 위한 spin column부터 dye-based one-step RT qPCR 시약, standard curve 작성을 위한 control template이 모두 포함되어 있는 all-in-one kit로 viral RNA의 copy수를 정량할 수 있다 (그림 1). 이를 사용해 lentivirus의 역가를 매우 빠르고 간편하면서도 정확하게 측정할 수 있다.

빠른 physical titer 측정으로 재현성 높은 실험 가능
Functional titer를 측정하는 일반적인 titration법은 형광 마커 유전자, 항생제 저항 유전자 또는 세포의 염색을 검출하는 방법으로 일반적으로2~10일 정도의 기간이 소요된다 (그림 2). 이 중 가장 빠른 방법인flow cytometry를 사용해 형광 마커 유전자를 검출하는 방법 역시 48시간 이상의 시간이 소요된다.
이렇게 functional titer를 측정하는 방법은 시간이 많이 소요되기 때문에 장기 보관 과정에서 반복적인 동결 및 용해 과정으로 lentivirus stock의 감염성이 감소하게 된다. 이러한 방법으로는 감소된 감염성 측정이 불가능하고 transduction 시 수일 전의 결과값으로 바탕으로 MOI를 계산하게 된다.

반면 Lenti-X™ qRT-PCR Titration Kit로 physical titer를 측정하게 되면 4시간 이내 역가를 측정하여 titration에 필요한 시간을 단축하기 때문에 바이러스의 harvest후 당일에 transduction을 진행할 수 있다. 따라서 lentivirus stock의 보관 과정에서 발생할 수 있는 감염성 저하 없이 정확한 MOI를 계산할 수 있고, 이를 통해 재현성 높은 결과를 얻을 수 있다.

qRT-PCR titration

Lenti-X™ qRT-PCR Titration Kit에는 qRT-PCR 시약이 포함되어 있으며, lentivirus의 backbone 서열 혹은 여기에 cloning된 유전자 (cDNA 및 shRNA 외)와 관계없이 모든 HIV-1 유래의 lentivirus에 적용이 가능하다. 또한 RNA 정제 단계에서 DNase I을 처리하기 때문에 lentivirus 생산 시 transfection한 plasmid DNA의 잔여물의 영향 없이 정확하게 정량할 수 있다.

패널 A. 2 X 109 ~ 2 X 102 범위 Lenti-X RNA Control Template을 이 제품을 사용한 qRT-PCR로 검출한 경우의 증폭곡선으로 106배 이상의 넓은 범위에서 titration이 가능하다는 것을 확인할 수 있다. 패널 B. 증폭곡선을 바탕으로 작성한 standard curve이다. 이때 Ct 값과 RNA copy수 (log scale)가 뛰어난 선형성을 보인다 (R2=1.00, PCR 효율 99.3%).

Lentivirus의 genome을 정제하여 10배씩 희석한 후 측정한 titration 결과이다. Lentivirus는 Lenti-X™ 시리즈의 시스템으로 생산하여 transfection 후 3일 뒤에 lentivirus의 상등액을 회수하여 사용하였고, 제작된 lentivirus를 -80℃에서 titration 전까지 보관 후 사용하였다.

바이러스의 감염성과 RNA titration의 상관관계

qRT-PCR을 사용한 titration kit는 사용이 편리하고 재현성이 높으며 짧은 시간 안에 정확한 결과를 제공하기 때문에 바이러스의 titration에 이상적이다. 또한 바이러스 stock의 functional titer (IFU/㎖)을 별도의 독립적인 방법으로 같이 측정하면 RNA titer (copy/㎖)와 functional titer의 계수 (IFU/Copy)를 구할 수 있다. 이 계수를 활용하면 추후 실험에서 짧은 시간내에 얻은 RNA titer를 활용해 functional titer (IFU/㎖)를 추정할 수 있고 이를 바탕으로 MOI를 계산할 수 있다.
