ICELL8 cx Single Cell System으로 분리된 single cell은 빈 5,184 nano well의 350V chip(Code 640019)으로 분주되고, CellSelect Software를 사용하여 single cell이 분주되어 있는 well을 식별, 선택한 후 SMART RT-PCR 시약을 선택한 well에 추가로 분주한다. single cell과 RT-PCR 시약이 분주된 chip은 ICELL8 cx Thermal cycler로 옮겨서 1st strand cDNA의 합성과 증폭을 위해 one-step RT-PCR이 이뤄지게 된다. 이렇게 준비된 full-length cDNA는 TDE1(Illumina Tagment DNA Enzyme)에 의해 tag가 지정되고, 이후 forward 및 reverse indexing primer(Code 640205 및 640218)를 사용하여 증폭된다. 이와 같이 제작된 library를 회수, 정제 및 증폭하여 최종 sequencing이 가능한 library를 준비할 수 있다.
