효과적인 상호작용 탐색을 위해서는 Matchmaker Gold Yeast Two-Hybrid System과 함께 Mate & Plate Libraries 사용을 강력하게 추천한다 (1). 이 제품은 cDNA library를 미리 yeast에 transformation시켜 library 증폭이나 별도의 transformation 과정없이 가장 간단한 방법으로 two-hybrid screening을 수행할 수 있다. Mate & Plate library는 cDNA library로부터 prey 단백질을 발현하는 Y187 MATαstrain과 bait 단백질을 발현하는 Y2HGold (MATa) reporter strain을 mating 하면 하나의 yeast에서 두 개의 단백질을 발현시킬 수 있다. 따라서 library를 가지고 있는 strain과 bait 단백질을 발현하는 strain을 하나의 tube에 넣고 배양한 후 선택배지에 도말하면 screening 과정을 시작할 수 있다 (Figure 1).
Normalized & Universal Libraries
Library normalization은 세포에서 상대적으로 다량 발현되는 cDNA를 선택적으로 제거하여 낮은 수준으로 발현된 cDNA의 상대적 함량을 높여 준다. 즉, housekeeping gene cDNA 등과 같이 상대적으로 풍부하게 존재하는 cDNA를 감소시켜 선별 과정에서 생성되는 false positive를 크게 감소시킨다 (1). 따라서 선별을 위한 더욱 다양한 수의 clone을 간편하게 확보할 수 있으며, 발현 수준이 낮은 단백질과의 상호작용을 검출할 가능성도 높아진다.

Universal Library는 가장 광범위한 유전자 발현을 대변할 수 있도록 전체 조직 (whole tissues)으로부터 제작이 되었으며, 역시 normalization 되어 있다(2). 따라서 목적 단백질과 상호작용하는 미지의 단백질을 발견할 가능성이 더욱 향상되었다.
