제품설명
Xfect Protein Transfection Reagent는 Clontech이 독자적으로 개발한 단백질용 transfection 시약이다. 목적 단백질이 세포막을 통과 할 수 있도록 세포 투과성 펩티드(CPP;cell-penetrating peptide)를 채용해 목적 단백질을 고효율로 세포로 도입할 수 있다. 또한 세포 독성이 낮고 단백질의 활성을 유지한 상태로 세포에 도입할 수 있다(그림 1). 본 제품은 기존의 단백질 도입 효율이 낮다고 알려진 human 유래의 부유 세포나 mouse 유래 ES세포 등에도 효과적이다. Plasmid DNA를 이용한 유전자 도입시 plasmid DNA로부터 mRNA 전사를 거쳐 목적 단백질이 세포내에서 발현 될 때까지 12~24시간을 필요로 하지만, 본 제품을 이용하면 활성이 유지된 단백질을 1~2시간만에 세포에 직접 도입하는 것이 가능하다.

Xfect Protein Transfection Reagent or Product C was used to transfect Jurkat cells with various amount of b-galactosidase (b-gal) according to the manufacturers'recommended protocol. One hour post-transfection, the cells were assayed for b-gal activity using Clontech’s Luminescent b-galactosidase Detection Kit II (Cat. No. 631712). Xfect Protein Transfection Reagent displayed a far higher signal for b-gal than did Product C. Panel B. HeLa cells transfected with 4 μg of b-gal demonstrated higher efficiency and a higher amount of b-gal protein per cell when transfection was performed with Xfect Protein Transfection Reagent (middle panel) than with Product C (righthand panel).

그림 4. Xfect Protein Transfection Reagent provides much higher cell viability than Product P when transfecting HeLa cells. HeLa cells were seeded in 96-well plate (3 x 104 cells per well) one day prior to transfection. The cells were transfected with 0.25 ㎍ of beta-galactosidase using Product P or Xfect Protein Transfection Reagent for 4 hours. A WST-1 proliferation assay was performed to assess cytotoxicity in the transfected samples relative to untreated HeLa cells.
