제품설명
RNAiso Plus는 동식물의 조직이나 배양 세포에서 간편하고 신속히 RNA를 분리할 수 있는 total RNA 추출 시약이다.
RNAiso Plus 용액을 첨가하여 조직이나 세포를 파쇄한 후, chloroform을 파쇄액에 첨가해 잘 혼합한 뒤 원심분리하면 3개의 층으로 분리된다. 이때 상부의 투명한 수층에 RNA가 있고, 반고체의 중간층에는 DNA, 마지막으로 적색*을 나타내는 하층의 유기용매 층에는 단백질, 다당류, 지방산, 세포의 debris와 소량의 DNA가 포함되어 있다. 여기서 상부의 투명한 수층만 채취하여 Isopropanol 침전으로 total RNA를 회수할 수 있다.
이 제품을 사용하면 약 1시간 이내에 total RNA 추출 전 과정을 완료할 수 있으며, 분리된 total RNA는 RT-PCR**, Northern blot 분석, mRNA의 분리 및 in vitro translation 반응 등에 이용할 수 있다.
* 본 제품에서는 유기용매층이 적색을 나타내기 때문에, 경계면의 구분이 쉬워 사용이 보다 편리하다.
** 정제된 RNA 샘플을 RT-PCR에 이용하는 경우 미량의 gDNA 혼입에도 결과에 큰 영향을 미치기 때문에, 사용 전에 Recombinant DNase I (RNase-free) (Code 2270A) 처리를 권장한다.
