  • Aureobasidin A 내성은 yeast one- & two-hybrid 연구에 이상적인 selectable marker이며 reporter이다.
Aureobasidin A (AbA)는 낮은 농도 (0.1~0.5 μg/ml)에서도 yeast에 치명적인 독성을 가지는 cyclic depsipeptide계의 항생제 (Figure 1)이다. 감수성 균종으로는 Saccharomyces cerevisiae, Schizosaccharomyces pombe, Candida glabrata, Aspergillus nidulans, A. niger 가 있다. AbA는 AUR1 유전자에 의해 발현되는 yeast 효소인 inositol phosphorylceramide (IPC) synthase를 억제한다 (1, 2). 형질전환된 yeast에서 돌연변이 유전자인 AUR1-C이 발현되면 yeast가 약제 내성을 나타내도록 하여 Matchmaker Gold Systems의 reporter로 작용한다.
Yeast One- & Two-Hybrid 연구를 위한 완벽한 Reporter
AbA는 내성이 없는 yeast를 단순히 생장 억제시키기 보다는 죽이기 때문에 AbA selection은 사실상 nutritional marker (예 : HIS3) 단독으로 사용하였을 경우 엄격하지 않은 screening 과정으로 인하여 발생되는 background colony를 제거시켜 준다. AbA 기반의 selection은 진짜 positive clone의 선별과 확인에 크게 도움이 된다. AbA selection 단독으로 사용하여 엄격하지 않은 일차 screening 과정에서 선별된 clone을 검사한 결과, 4 개의 모든 Matchmaker Gold reporter (AUR1-C, HIS3, ADE2, MEL1)를 발현하는 진짜 positive가 높은 비율로 확인되었다. 좀 더 많은 정보는 카탈로그 Matchmaker Gold Yeast Two-Hybrid System 부분에서 볼 수 있다.
Figure 1. Structure of Aureobasidin A. Aureobasidin A (MW 1,100) is a cyclic depsipeptide antibiotic isolated from the fungus, Aureobasidium pullulans R106. AbA inhibits the product of the yeast AUR1 gene (inositol phosphorylceramide synthase) and is toxic to S. cerevisiae at low concentrations (0.1 μg/ml). A dominant mutant allele, AUR1-C, confers resistance to AbA, and its expression can be used as a selectable marker.
