NOTE: We recommend using collagen-coated plates or flasks for efficient culturing of frozen stocks. Vessels coated with compounds other than collagen may also provide suitable growth substrates (e.g. poly-L-lysine), but only collagen-coated plates (e.g. BD BioCoat Cellware, Collagen Type I) have been tested at Clontech. Once recovered, the cells may be cultured directly on tissue culture plastic. However, if adherence is poor, we recommend using only collagen-coated vessels.

Tet-On Advanced and Tet-On inducible cell lines are host cell lines that greatly simplify the task of setting up a tetracycline-inducible gene expression system that is activated by adding doxycycline (Dox) to the culture medium (Figure 1; 1, 2). Our Tet-Off¢ç Advanced and Tet-Off inducible cell lines are also available, which are activated by removing Dox from the culture medium.
These rigorously tested clonal cell lines:
  • Stably express either the second-generation Tet-On Advanced transactivator protein or the original Tet-On transactivator protein
  • Allow you to induce expression of your gene-of-interest by adding doxycycline (Dox) to the culture of transfected cells
  • Can save you up to two months of time in developing a complete Tet-controlled system.

Easily create an inducible system by first placing your gene under control of a tetracycline-responsive promoter (e.g. PTight in pTRE-Tight), transfecting it into one of these G418-resistant cell lines, and selecting for double-stable clones.
