제품 설명
AAV Rapid Titer ELISA Kits는 AAV 2, 8, 9 serotype에 대한 capsid 단백질을 검출하고 정량함으로써 AAV의 역가를 효율적으로 측정하게 된다. 이 분석법은 virus capsid 단백질에 특이적으로 결합하는 capture antibody를 사용하여 AAV 입자에 대한 높은 특이성을 보장한다. AAV Rapid Titer ELISA Kit의 프로토콜은 한 번의 washing 단계로 빠르고 편리하게 정량할 수 있으며, hands-on time을 최소화한다.

① ELISA plate에 미리 코팅된 anti-AAV capture antibody를 이용하여 샘플 또는 AAV standard 물질을 결합한다 (실온에서 30분간 incubation).
② AAV HRP-conjugated detection antibody를 plate에 첨가한 후, 실온에서 30분간 더 incubation하고 1회 washing한다.
③ TMB 기질을 첨가하여 15분간 incubation한다.
④ 정지 용액으로 반응을 중단시키고 신호를 판독하며, 샘플 흡광도 값을 표준 곡선과 비교하여 정량한다.

standard curve는 제품 내 구성품인 동결건조된 AAV2 (Panel A), AAV8 (Panel B), AAV9 (Panel C) Empty Particle Control의 희석액을 사용하여 AAV Rapid Titer ELISA Kits (Single Wash)로 분석하였다. 표준 곡선의 R² 값 (점선)은 선형 회귀 분석을 기반으로 한다. Panel D. 검출 한계 (LOD, limits of detection) 및 정량 한계 (LOQ, limits of quantification) 를 나타낸 표
