기작

Doxycycline으로 조절되는 ”transactivators”(Tet-On과 Tet-Off Advanced)는 3 개의 작은 HSV VP16 단백질의 minimal transcription activation domain과 TetR-유래의 DNA-binding domain이 연결된 융합 단백질이다.
Transactivators는 높은 특이성으로 Tetracycline Response Element (TRE)와 결합한다. TRE는 PLVX-Tight-Puro의 PTight promoter에 위치하고 있으며 실험자의 목적 유전자를 포함한다. 시스템이 유도되면 PTight 로부터 목적 유전자가 높은 수준으로 전사된다.
Tet-Off Advanced System은 Dox가 있을 때 basal state가 유지되고, Dox가 제거되면 전사가 진행 된다. 반대로 Tet-On Advanced System은 Dox가 있을 때 전사가 활성화된다 (Figure 1).

Figure 1. The Lenti-X Tet-Advanced Systems are highly inducible. Using equal amounts of high-titer supernatants, HeLa cells cultured at the indicated concentrations of Dox were cotransduced for 8 hr with an LVXTight-Puro-Luc lentivirus and an LVX-Tet-On Advanced lentivirus. Cultures were harvested after 48 hr and assayed for luciferase activity. Luciferase was expressed at very high levels, while the basal/uninduced expression level was very low.
