Adeno-X Rapid Titer Kit
Adeno-X Rapid Titer Kit는 항체를 이용하여 1 세대 E1-deleted adenovirus역가를 간단하고 신속하게 측정할 수 있다 (2). 본 제품은 감염된 세포를 hexon 특이적 항체로 염색하는 방법을 기반으로 하고 있다. Hexon 단백질은 adenoviral genome으로 유도되며 adenovirus 복제에 필요한 adenoviral capsid의 필수 구성 요소이다. 그러나 hexon 단백질의 발현은 E1 유전자 산물에 의존적이다. 따라서 감염된 세포만이 hexon 단백질을 생산하기 때문에 HEK293과 같은 E1 trans-complementing 세포 (3)는 감염된 활성을 측정하는데 사용될 수 있다. Rapid Titer 검출법은 실험 준비을 위하여 1~2 시간, 그리고 2 일 후 감염된 세포를 표식, 염색하고 측정하는데 추가 3 시간 정도 소요된다. 각 염색된 세포는 single infection unit와 일치한다 (Figure 2).
Figure 2. With the Adeno-X Rapid Titer Kit, infected cells turn brown, so they are easily counted using a microscope. Healthy HEK 293 cells (5 x 105/ml) were added to each well (1 ml/well) of a 12-well culture plate and then infected with various dilutions of an adenovirus stock according to the Adeno-X Rapid Titer Protocol. Following a 48 hr incubation, infected cells were identified by adding mouse anti-hexon antibody, HRP-conjugated rat anti-mouse antibody, and DAB substrate. This microscope image, taken with a 20X objective, shows a field of infected cells in a well that received a 10-5 viral dilution.
